The compound light microscope is frequently employed for observing transparent objects that can be accommodated on a microscope slide. A microscope slide is a slender, rectangular glass plate capable of accommodating a diverse range of microscopic specimens.
Microscope slides offer optimal optical clarity, facilitating precise diagnostic assessments. The reduction of impurities, including metallic contaminants in common float glass, minimizes fluorescence and ensures a uniform platform for sample observation.

Originally, a microscope slide, or 'slider,' was crafted from ivory or bone, containing specimens secured between disks of transparent mica. These gained popularity in Victorian England until the Royal Microscopical Society introduced the standardized microscope slide—a thin sheet of glass designed to hold objects for examination under a microscope. The standard dimensions for a microscope slide are 75 x 25 mm (3" x 1") with a thickness of about 1.0 mm. Various other sizes are also available to cater to specific needs.
Given that high power microscopes have a limited focal region, the specimen is typically positioned in the middle of the slide, with a much thinner square, circle, or rectangle of glass placed over it. This smaller glass sheet is known as a cover slip or cover glass, measuring between 18 and 25 mm on each side and 0.085 to 0.25 mm in thickness. The cover glass serves two essential purposes: protecting the microscope's objective lens from direct contact with the specimen and ensuring an even thickness (in wet mounts) for optimal viewing. The precise thickness of the coverslip is crucial for achieving high-resolution microscopy.

What is the Difference between Prepared Slides and Wet Mounts?

A wet mount slide involves the use of a liquid drop, typically containing a fresh or live specimen, and is designed for temporary observation. In contrast, a prepared slide has already undergone preparation, often involving the application of chemicals to preserve and permanently mount both the specimen and the coverslip.
